Will this work with a backpack?

Yes it can. What we think will make the most sense is, to wear the pack first, and the backpack above it. We assume you will want to keep the bino pack on longer than the backpack, that's why it should come first. Also, you may consider removing/relocating the detachable pouches from the top of the straps and either use them on other molle type bag (if your backpack for example is using the molle system), or tuck them away for the next time. I’m a big man, how big of man will these fit? I’m 6’1” and weigh 375 pounds.

Hi there and thanks for your question! I am 6’2” and 205 and there is a LOT of room on the straps for me. This pack was built to fit most sizes, even big sizes. We can not estimate specific weight as it also has to do with body shapes. I suggest if you want to give it a try, order it and check it out. Does this harness/case have Molle straps on bottom of main compartment?

Do the straps interfere with sholdering a rifle?

You can take off the pockets for the straps. I haven’t had an issue with it at all.
