Will galaxy a12 that already has a screen protector and silicone case that stretches over the back of the phone fit in this leather wallet

Maybe not. The case is a very snug fit on my phone so unless the silicon case is very very thin it is unlikely it will fit.

Would this fit my A21s?

This case is for the A12. The A21 is the same size so would fit but at the back there is a window for the camera lenses to show through. The lenses for the A21 are a different shape/configuration so may not show through properly. I would think that iCatchy make one for the A21.

Does this lock the phone when closed?

No it doesn't, but I lock mine before folding, it's brilliant, space for cards and cash notes
