Will this work on a 2 stroke

Yes. I use it on my 2002 Merc. 50hp 3 cylinder two-stroke without any issues

Will this work for an inline 6 cylinder on a 65 mustang, need to replace the distributor?

This is perfect for your job. Just make sure you have it on top dead center for #1 cylinder. Remove #1 plug, place finger over spark plug hole, slowly rotate engine until you feel pressure on finger, watch timing mark until on zero. Then you can remove distributer and replace it with a new MSD. Better spark and more reliable.
