how to install self drilling drywall anchors?

Just run them in using a phillips screwdriver or a cordless drill/driver with phillips insert bit. A pilot hole makes starting them easier, and be careful to avoid studs. Be careful when running the supplied phillips screw into the anchor, they're a bit too big and can tear the plastic anchor loose from the drywall.

Will these hold with a number 6 drywall screw?

They are designed for and come with number 8 wood screws. The 6 drywall may be loose but would go in. Depends on your project.

can these drill through a stud?

If you are drilling directly into a stud you do not need to use the plug. The plug is for walls that will not support the weight of a shelf or heavy object with just a screw and need the plug to hold the screw in place. You need to use a drill bit to make the hole and screw the plug in if there is no stud to support the screw alone. Then the screw can go through the bracket into the plug.

Will this fit a 1/4"-20 X 1-1/4" screw? I can not find something to match this size. Thank you

I don't think so. The screws that come with it are #8 x 1-1/4'', and #8 is generally described as 5/32, which is considerably smaller than 1/4 (which would be 8/32).
