Q: How many pencils/pens will this hold? Also, will the stitching hold up?

It has 7 pencil slots but in back of them is a pouch area where you can fit many more pens, you'll want them to have a clip to hold them in place. Also, the pen slots don't go all the way across, there is a pouch there that I keep a ruler in. The stitching is very good and will hold up. The quality is good and you can expect this to last many years. I really like mine and carry it with me everywhere as part of my sketch kit.

Q: Do you think a high lighter would fit the small individual areas or at least fit 4 high lighters in the larger pocket?

Depends on the brand of highlighter, Some Sharpie may fit the "small individual areas", on mine a pencil (pencils) fits nicely, as will an 'Extra Fine Point Permanent Marker', the larger pocket may fit 4 smaller high-lighters.

Q: Would this work for cosmetic brushes?

This would work for cosmetic brushes and it will protect them as well. The leather is is supple but holds it shape. I use mine daily it is one of my favorite thing.

Q: Can it fit a calculator in it?

I bought one, and yes a calculator will fit. Not necessarily in the left inside pocket, but behind it. I have all sorts of things crammed in mine.
