What is the clear rubber white thing on the top of the gun?

It's for a vacuum sealer hose. This smoking gun is sold under different labels with different accessories. If you check out the COSTWAY 2-in-1 Smoker Gun or the JoChef, you'll see that the gun is the same as this one, but the product comes with a vacuum searler hose and some bags.

Can you easily remove the batteries?

No! Pretty certain the batteries are not meant to be removed. The smoker is charged thru a USB port.

Would this work as a vehicle vacuum leak detector??

I don't see why it wouldn't, all you are doing is putting smoke in the engine bay and looking to see where it might get sucked in. I think it would be very good. JMHO
